Construction and Validation of a Novel Prognostic Signature for Intestinal Type of Gastric Cancer

Background: Intestinal type of gastric cancer (IGC) is the largest subtype (GC). The purpose was to construct a prognostic signature for IGC patients improve accuracy. Methods: microarray datasets and associated clinical characteristics HGD were obtained from Gene Expression Omnibus (GEO) database. Based on differential expression analysis between IGC, related genes (DEGs) identified in training group by univariate COX regression analysis. least absolute shrinkage selection operator (LASSO) used an optimal signature. enrichment performed using Set Enrichment Analysis (GSEA). performance nomogram assessed calibration curve concordance index (C-index). results validated testing group. Findings: We 35 DGEs cohort. nine-gene established LASSO independent risk factor both cohorts. In GSEA analysis, gene high-risk enriched hedgehog signaling, epithelial mesenchymal transition, angiogenesis. showed good with C-index 0.81 (95%CI: 0.76-0.86) 0.70 0.63-0.77) cohorts, respectively. Interpretation: verified prediction patients, which might identify subgroups select more suitable therapeutic options.
